Time-Resolved Optical Tomography in Preclinical Studies: Propagation of Excitation and Fluorescence Photons
نویسندگان
چکیده
We present time-resolved methods that rely on near-infrared photons to image the optical properties and distribution of fluorescent probes in small laboratory animals. The scanner assembled in our laboratory is described. The coupled diffusion equations of excitation and fluorescence photons in highly scattering tissues were solved using the three-dimensional Finite Element Method (FEM) provided by Comsol. The computed results allowed to yield photon density maps and the temporal profiles of photons. Simulations were carried-out on two test objects: a resin cylinder and a mouse phantom. The latter required taking into account the straight propagation of photons between the optical fibers and the animal phantom. The results demonstrate the potential applications of the method to pre-clinical imaging.

Superfluorescence appears as an intense background in femtosecond time-resolved fluorescence noncollinear optical parametric amplification spectroscopy, which severely interferes the reliable acquisition of the time-resolved fluorescence spectra especially for an optically dilute sample. Superfluorescence originates from the optical amplification of the vacuum quantum noise, which would be inev...

متن کاملMonte Carlo based method for fluorescence tomographic imaging with lifetime multiplexing using time gates
Time-resolved fluorescence optical tomography allows 3-dimensional localization of multiple fluorophores based on lifetime contrast while providing a unique data set for improved resolution. However, to employ the full fluorescence time measurements, a light propagation model that accurately simulates weakly diffused and multiple scattered photons is required. In this article, we derive a compu...
متن کاملImprovement of fluorescence-enhanced optical tomography with improved optical filtering and accurate model-based reconstruction algorithms.
The goal of preclinical fluorescence-enhanced optical tomography (FEOT) is to provide three-dimensional fluorophore distribution for a myriad of drug and disease discovery studies in small animals. Effective measurements, as well as fast and robust image reconstruction, are necessary for extensive applications. Compared to bioluminescence tomography (BLT), FEOT may result in improved image qual...
